Dear colleagues,
For a research project, we need to introduce a reliable method for the
determination of ACE genotype (insertion/deletion polymorphism, intron
16).
Having screened the literature, I note increasing concern about
potential mistyping mostly affecting I/D heterozygotes.
Can anyone recommend a PCR protocol which is less prone to mistyping
than the original (and widely used) method by Rigat (Nucl Acids Res
1992;20:1433).
Should all analyses be done in duplicate using two different approaches
(confirmatory PCR)?
Thank you for your input.
