Hi,
When we do prednisolone absorption tests the prednisolone peaks and is on its way down within 200 minutes. I would say 24 hours would be more than adequate but you could perhaps get away with 12. However, some patients are slow absorbers and it may take longer.
We offer a mass spec service for this situation where we can measure cortisol (and prednisolone) if you want to send a sample here.

Subject: Prednisolone
Does any one have any guidance on how long a patient should be off prednisolone before cortisol can be measured accurately. We are using the Roche modular cortisol assay.
