I think you only use the hotspot thing in the multislice view. You
can change the slices to coronal for multislice view though, in the
options (but not sagittal for some reason).

You can do this with ROIs in MRIcro, if you define an ROI for each
point activated, and then use the ROI comparison menu. You can't do
this kind of thing with the hotspots feature.
To get an idea for the colours corresponding to the 24 bit colour
numbers, you could make a chart using a little program.
Here's one in matlab:
