We offer an assay for the measurement of Vitamin C in heparinised
plasma by HPLC with fluorescent detection. Ascorbate is oxidised
by iodine to dehydroascorbate which is coupled to a fluorescent
agent. Samples can be stabilised by adding the plasma to 5%
metaphosphoric acid and then frozen. There is no loss in vitamin C
for up to 4 hours prior to blood separation and treatment. However,
samples should be separated as soon as possible as in patients
where there is increased free radical activity Vit C is less stable even
in-vitro (Schorah et al., Am J Clin Nutr 1996; 63: 760-5). CRP needs
to be measured at the same time because of the negative
correlation with the inflammatory response.
