We run the Microgenics CEDIA Gentamicin assay on an Olympus AU640 using a 20% detergent solution (Extran or Decon are suitable) for reagent probe washing according to Olympus protocol:
Contamination Parameters AU640
Issued 11-02-03
Contamination
No. Preceding Following Reagent Probe Wash Cancel Mixer Cuvette
Test Name Test Name Cleaner Kind Count
5. All Gent 420 CLN 20% Extran* 5 No Y Y
The assay requires calibration approximately every 4 days (2 point)and internal QC run with around 4.6% cv.
Between run imprecision is actually considerably better than this, however the QC tend drift downward over time (in common with other CEDIA assays on this system), hence the need to pull the assay back into line at around the 4 day mark.

Subject: Gentamicin assays
Is anyone successfully running a gentamicin assay on Olympus 640 or 400
please?
If so, what protocol are you following to avoid contamination with the
gentamicin present in many Olympus reagents? Any recent experience, good or
bad, with reagents from Olympus or other supplier would be of interest.
Many thanks
