Preparation of Material
Sodium fluoride solution 4g/L (supplied by BDH Laboratory supplies, Poole,
Dorset Cat number:10246 4T.)
DGlucose solution 36g/L (supplied by BDH Laboratory supplies, Poole, Dorset
Cat number 1.08337.0250)
Aseptically collected defibrinated horse blood (E & O Laboratories Ltd). .
Sample Preparation - (Done the day before distribution)
During these stages, wear latex disposable gloves.
To a 100ml bottle of horse blood, add 5.0ml sodium fluoride. The glucose
level is then adjusted to the required level (different each circulation) by
adding varying amounts of glucose solution.
The following is an approximate guide:
Final Level Amount Added
1.0mmol/L 0.5ml
2.0mmol/L 1.0ml
5.0mmol/L 2.5ml
10.0mmol/L 6.0ml
15.0mmol/L 9.0ml
20.0mmol/L 12.5ml
Allow to mix for 30 minutes on roller mixer then assay using MediSense
Glucose Meter. Adjust value by further addition of glucose solution if
necessary. Re-mix then test again.
