Dear CCPEM Community,
I hope this email finds you well. I am currently working on
integrating Relion 5 for tomography in Scipion and as I navigate
through this process, I have encountered several questions and
challenges that I believe your expertise can help address.
I kindly request your assistance in understanding and resolving the
following issues:
1. Is there a way to get the projections matrices that can be used to
project the 3D particles on the tilt-images, just as they were written
in Relion 4 in the file called tomograms.star (fields _rlnTomoProjX
#1, _rlnTomoProjY #2, _rlnTomoProjZ #3, _rlnTomoProjW #4? If not, is
there a way to calculate them using the data contained in the star
files managed by Relion5?
2. Regarding the EDF file, I'd like to know if it's used for something
after the tilt-series alignment as it is referenced in the
tomograms.star file generated in the Tomogram reconstruction step.
3. Can the pseudo-subtomograms from a processing made using Relion 4
be used to work directly with Relion 5 or should some adaptations be
required?
4. I've been exploring the advanced arguments of the program
relion_tomo_subtomo_mpi, and I've observed that the particles can be
generated non-premultiplied by the CTF. Does it affect both 2D and 3D
particle representations? In the affirmative case, is it possible to
continue the processing in Relion5 with the non-CTF-pre-multiplied
particles?
