DOGTAG – A GENETICALLY ENCODED PROXIMITY LABELLING STRATEGY TO CAPTURE
PROBLEMATIC PROTEIN-PROTEIN INTERACTIONS
Capturing protein-protein interactions is key to understanding
biological processes yet many interactions are missed due to being weak,
transient or not amenable to heterologous or biochemical examination.
There are many existing strategies to investigate protein-protein
interactions (e.g. BioID, Y-2-H, IP-MS) but none allow for naïve,
non-invasive, in-vivo examination of weak or transient protein
interactions under physiologically relevant conditions. We are piloting
a novel genetically encoded method allowing for proximity labelling
experiments to be performed unaffected by these limitations. The aim of
the project is to develop the method and ultimately perform differential
discovery proteomic screens to identify enzyme-substrate interactions in
whole organisms under biologically relevant stimuli.
Summary of Skills, Experience and Qualifications
Applicants should have, or be about to obtain, a PhD, and have a solid
background in molecular biology and biochemistry. Experience in
proteomic experimental planning and data analysis would be useful.
Experience of methods development, working with membrane proteins,
protein biochemistry, proximity labelling screens and proteomic sample
processing would be advantageous.
Appointment as a Postdoctoral Research Assistant on the Grade 7 salary
scale (£31,604 - £38,833) is dependent upon you having been awarded a
PhD. An appointment may be considered if you are shortly expected to be
awarded a PhD in which case the initial appointment will be made as a
Research Assistant on the Training Grade 7 salary scale (Spinal Point
28, £30,688).
