Dear CCP4 community,
I would like some advise on SEC columns. We have the "AKTA Purifier 10" FPLC system from GE, and use the "Superdex 200 10/300 GL" column for all SEC work. I have recently heard there are other columns that are better suited to SAXS sample prep. Is this true, and if so what column do you use?
One of the biggest concerns with SAXS sample prep is ensuring the sample and buffer blank are identical, so that subtraction will be accurate. I was told that a good strategy is to use the buffer from the SEC run, after about 1 CV has ran through after the protein. Has anyone had success with this as a final step, or should dialysis follow?
I appreciate any advise that is offered, and thank you in advance! The "Shodex KW-403-4F" column (HPLC) was recommended to me, but we only have the GE Purifier 10 system and I don't think they are compatible.
