Hi,
You can manually estimate the filter, but there is no "correct" answer and it is likely that your estimate will be very conservative. The estimates given by the GUI or cutoffcalc are likely to be less conservative and therefore give you a _little_ more statistical power. Typically it doesn't matter a great deal though. I would go with one of the estimates that you get from the GUI or cutoffcalc (if you are concerned about the difference, then choose the larger one to be slightly conservative).
