Dear Mona,
it looks like that the SPM.mat file, your are calling, does not belong to the spmT images, which should be in the same folder (both *.hdr and *.img). So, you should check whether the belong together or not. My guess from the error message is, that they do not belong to each other. You may want to display that spmT_0002.img to check, which dimension it has, and check it against the values, given in SPM.xCon.Vspm
With respect to the other question: It should work in the same way as in SPM2. You probably should check your design (with 'Review' from the main panel, or 'Design' in the result window) to make sure that the groups are in the right columns of your design matrix and thus matching your contrast.
