No this is not the right format. You should provide an image (img or
nii) with a mask corresponding to your prior and then SPM will
generate the pQ matrix that you'll be able to use the next time. See
section 37.7.2 (page 406) of the manual. In principle it should be
possible to specify priors as a list of coordinates but this is not
implemented yet.
