Hi Alexander,
no, it's not required, but this is preferable (especially to put a prefix such as con_ and pat_) as it will make your design.mat easier to make.
More generally, all your TBSS (and VBM) images ready to be analysed are concatenated following the alphabetical order, so your statistical design have to match it as well.
If you have already run all the processing steps of TBSS, you just need to adjust your design.mat to match the order given by a "ls *_to_target.nii.gz" in your FA directory.
