Hi Mirko,
I'm afraid I don't have time to test this right now, but I think it
should work if you use imcalc but when selecting the files change the
frames selector (lower right of the bit above the files list) from 1
to something like 1:100 (or however many frames/volumes are in this 4D
NIfTI). You should then be able to select the multiple frames, and
enter an expression as usual. (BTW, you must be using SPM5 to work
with multi-frame NIfTI volumes like this, I believe)
If you do have many frames, you will probably want to use imcalc with
the dmtx flag, and the expression 'sum(X)', instead of entering
'i1+i2+...'.
