MMBGX/MMseq: Modelling isoform expression and differential splicing
from Exon array and
RNA-seq data
Abstract
Splicing is the process by which introns in pre-mRNA transcripts are
removed and the remaining exons connected to form mature mRNAs. Genes
with more than two exons may produce multiple versions of mRNA, known
as isoforms, made up of different combinations of exons. Alternative
splicing is thus an important source of diversity in proteins, and is
expected to be important in normal development and disease.
Most approaches have so far focused on detecting changes in exon
inclusion rates instead of the expression of the isoforms themselves.
Here, we present two statistical approaches, MMBGX and MMseq, for
estimating Ensembl isoform expression using Affymetrix Exon arrays and
RNA-seq data respectively.
