Short Summary: Two studies were used to evaluate how nalmefene is used in the management of alcohol dependence in France. After 1 year, new nalmefene users had substantially reduced alcohol consumption. Insurance data confirmed external validity of the population sample and the observation that patients often discontinue treatment over 1 year.

Short Summary: Two studies were used to evaluate the safety and persistence of nalmefene in the routine management of alcohol dependence in Europe as part of the overall risk management plan. Although adverse events appeared more common in over 65-year olds, there were no major differences in safety outcomes between the total population, and those with psychiatric and somatic comorbidities or those with a history of seizures. Findings were in line with those from previous clinical studies. The differing rates of persistence beyond 1 year (49% in the prospective study and < 5% in the multi-database retrospective cohort study) likely reflect the different study methodologies and serve to highlight the relevance of follow-up support.

Short Summary: Intranasal oxytocin did not significantly reduce alcohol craving or relapse compared with placebo the first four weeks after detoxification in alcohol-dependent patients. The oxytocin treatment group did report a statistically significant decrease in self-reported nervousness.

Short Summary: To examine predictive validity of the Addictions Neuroclinical Assessment negative emotionality domain, we assessed longitudinal associations among this domain, coping motives and drinking intensity among treatment seekers who experienced a lapse to drinking. Negative emotionality was indirectly associated with drinking intensity following treatment initiation through coping motives for alcohol use.

Short Summary: Female central amygdala (CeA) GABAA receptor current kinetics are faster than males and depend on estrous cycle, with alcohol dependent animals having slower kinetics. Acute alcohol increases presynaptic GABA release in male CeA but has no effect in naïve female CeA. High concentrations of alcohol increase GABA release in dependent female CeA.

Short Summary: This study shows that subjective intoxication, hangover severity and hangover frequency decline with age. The relationship between age and hangover severity is strongly mediated by subjective intoxication. An age-related decline in sensitivity to pain may in part explain the observed negative relationships.

Short Summary: This study confirms, for the first time in female rats, that RYGB increases and SG decreases alcohol intake. An experimental surgery, combining RYGB and SG, confirmed that removal of the remnant stomach decreases ethanol drinking. These results point to ghrelin as a possible contributor to increased ethanol consumption following RYGB.

 
 
Changes in the Gut Microbiota Following Bariatric Surgery Are Associated with Increased Alcohol Intake in a Female Rat Model

Short summary: The gut microbiota diversity and composition are altered in distinct ways after RYGB or VSG surgeries. We observed that a decrease in gut microbiota diversity is associated with EtOH intake after surgery and this may contribute to the increased incidence of AUD observed clinically in the bariatric surgery population.
