Alice

In P2 and P21?
Eleanor

Do you have the beta angle for your P21 cell?

Yes we have tried SHELXD

Hmm - pseudo translation shouldnt matter for finding ha sites..
of course your SG could be wrong - a translation of 0.33 0.5 0.72 means you will have absences along 0 k 0 whether the SG is P2 or P21
I presume you have tried SHELXD?

Hi all,

We are trying to phase a structure using experimental phasing (few different heavy atoms showing anomalous signal to 4-5A at best (otherwise 6-7A)) and native data are about 2.8-3.0A resolution. There is no homologous model for molecular replacement and Xtriage clearly showed the presence of a pseudo-translation.
The crystallographic info is:
Space group P21, cell parameters: 77,134,84,90,100,90
Pseudo-translation: 0.33 0.5 0.72

So far, we did not manage to find the sites.

How are the current softwares dealing with pseudo-translation for experimental phasing?
Can you point us to some structures solved by experimental phasing with a pseudo-translation? Especially how it was solved?

Thank you

Kamel and Alice
