You can generate the template using as many or as few subjects as you like; however, if you have multiple groups and your template contains only subjects from one group, a reviewer will likely raise an issue.

There are two elements to DARTEL:
(1) Template generation;
(2) Creation of the deformation fields.

The later allows you to add more subjects to the analysis without having to regenerate the template set again.

So if I use a DARTEL template for preprocessing of functional and anatomical data, I assume I will have to make sure that the subjects that are included in preprocessing are exactly the same ones whose anatomical images are used for creating the DARTEL template? Template_6 is not in MNI space, which is why the DARTEL needs to be normalized to MNI space for all analyses.

When you apply the deformations, make sure that your functional data are aligned to the structural images that were used for DARTEL.

I intend to create a DARTEL template (gm and wm) in SPM12 based on a good number of experimental subjects so as to replace the MNI template with the DARTEL template in preprocessing of the fMRI data (for univariate analysis). In this case, I assume I just need to do 'Run DARTEL template' but not the 'Normalize to MNI space' step (which is required to do for a VBM analysis)? My understanding is that the Template_6.nii is already in MNI space after 'Run DARTEL Template'. Also in my preprocessing analysis, in the 'normalization' step, I will apply the DARTEL template (deformation parameters) to each subject's images anyway.
