In the fMRI data, if there are two condition than the design matrix X will have the 2 columns. For e.g.,  in the SPM auditory data set, design matrix X has two columns because it has only two conditions. Thus, number of b's would be 2 only. but HRF has more number of taps than 2.

Than how the b would be the amplitude of HRF in that case?
Should i form the new design matrix (which should have more number of columns)?
