I don't think I understand your question.  The contrast you specified is the contrast for comparing the groups.  You would run [1 -1] to test males>females and [-1 1] for females<males.

Since I'm really interested to investigate the difference of two group, Can I define the contrasts like this?[1 0],[0 1] and then find the significant pvalues for each of them separately and remove the common pvalues' index, then consider the rest of each pvalues as effect of male or female? Does it seem reasonable way to find the difference of two group?
