No. Intensity drifts over time would be modelled by the high-pass filter in the GLM, which would fit a few cosine basis functions to the data, in addition to the various other effects that would typically be included in the design matrix. Alternatively, if you just want to model out linear effects with time, you could disable the high-pass filtering and include an additional covariate of (1:N)-N/2 in the design matrix.
