Dear Catherine Basically, this depends on how much your interpretation rests on the "expected" area and how specific this expectation is. If your peak is considerably off and/or your expectation was anatomically very specific, you basically need to conclude that your task did not work to activate this region. Hence you shouldn't continue with further analysis. If your hypothesis was more functional in the first case, you should go on based on the functional peak, considering that the inter-individual spead of local maxima can be >10 mm for some regions. In this case, it is acceptable to select the coordinate that is not the peak voxel (which empirically & anatomically makes more sense) as my functional ROI and compare mean beta weights for different conditions?
