Unequal sample sizes do not effect the mean difference between the groups. Thus, the contrast should be 1 -1.

For unequal sample sizes, the correction is done for the variance based on the number of subjects. Just select unequal variance.

Dear SPM users,

This is certainly a simple question for an expert.

I have two groups. One with 23 subjects and the other with 11 subjects.

I want to compare both groups with a second-level analysis (t-test, unpaired two samples).

What is the correct vector contrast

1 -1

or

1 -23/11

(to compensate for the unequal sizes of the groups)?

Thanks for your help,
