i have a doubt, HOW can i create ROIs on SPM?'
SPM isn't really designed to do this, except for writing the result of an analysis as an ROI. You'd better use software like MRIcro or Analyze for this.
and should i make that roi's on a template subject, or i must make on each subject of my study?'

Designing a protocol, testing for intrarater reliability, and then drawing on each subject of your study would be the gold standard - but may be hard to achieve if you have large numbers of subjects and/or regions.
There really isn't a "template subject" (there is one subject whose MRI has been brought into MNI space, but like every single brain it has idiosyncrasies making it non-representative).
Drawing on the actual templates which are averages built from many subjects is rarely an option as you will find them too blurry to define many boundaries, particularly cortical ones. Depending on your scientific question (and on your imaging method!) this might of course still be sufficient.
The accuracy of many of the anatomical approaches has been compared (in a slightly different context) in Heckemann RA et al. Neuroimage 2006.
