Weikai, You might look up these references: A. Lustig et al. "Density determination by analytical ultracentrifugation in a rapid dynamical gradient: application to lipid and detergent aggregates containing proteins." Biochim. Biophys. Acta 1115 (1991) 85-95. Ariel Lustig et al. "Molecular weight determination of membrane proteins by sedimentation equilibrium at the sucrose or Nycodenz- adjusted density of the hydrated detergent micelle." Biochim. Biophys. Acta 1464 (2000) 199-206 Ariel Lustig, a master of the old model E and the new Beckman instrument, did a lot of work for Jurg Rosenbusch just on this subject. One other aspect that needs to be addressed is the compressibility of the detergent micelles. This can vary from detergent to detergent. You can find much of what you want by delving into the surfactant/colloidal science literature. Also look up the work of Marc le Maire, who has studied detergent- protein interactions for years. If you can, get a copy of Michael Grabo's thesis (Solubilization and Reconstitution of Membrane Proteins, Ph. D. Thesis 1982. University of Basel, Switzerland); there are many useful nuggets in this work. Michael **************************************************************** R.
