This is really a question for GE or a Diffusion group (e.g., the ISMRM Diffusion-Perfusion study group), not SPM.  I will send you some ideas off-list, and any other interested listeners can contact me directly.  I believe SPM has a Diffusion toolbox, but I am not familiar with it (perhaps someone else can chime in here).  I have not read Susumu Mori's new book "Introduction to Diffusion Tensor Imaging" (Elsevier, 2007), but it seems to have some discussion about practical aspects of DTI measurements.

Subject: [SPM] New to DTI and tractography

Dear SPM'ers,

I am new to DTI and need some information about it. We are using a 3T GE MRI scanner and let's say we want to reduce the scan time to less 15 mins...to look at tracts in the frontal regions. What would be the best pulse sequence?

Also, can anyone tell me what (ideal) software and hardware for DTI and tractography would I need?
