Not necessarily. We have recently collated a number of databases distributed
across various servers into a single database and we had to ensure that
people remained opted out where we had duplicate entries for them.

But when you collect their details for the second time, in any event 
you'll only be able to use them for marketing if the person opts-in. 
Which is their choice, at the time.
