Title: Univariate and bivariate methodology for estimation
after a sequential clinical trial
Abstract:
A clinical trial recruits patients over a period of time, and the results
accumulate steadily throughout its duration. Yet, according to traditional
statistical methods, the size of the trial sample should
be determined in advance, and data collected on all subjects before
analysis proceeds. For ethical and economic reasons, the technique of
sequential testing has been developed in which the data
are examined at a series of interim analyses. The aim is to provide a
valid analysis as soon as sufficient evidence to reach a firm conclusion
has been obtained.
The analysis of any clinical trial involves the calculation of summary
information about the results. In a frequentist analysis, P-values and
point and interval estimates of the treatment effect are
usually given. In addition, one or more functions of the original
parameters, besides the treatment difference may be of interest.
Traditional methodology based on the frequentist interpretation of
probability must be reinterpreted to fit within the sequential framework.
This talk will consider novel methods of analysis valid after a study has
been monitored sequentially. Application to both univariate and bivariate
patient responses will be developed. The
approaches will be compared with a traditional fixed sample size analysis,
their properties investigated and their accuracy demonstrated.
