I have been processing some virus cryo-ET data. So far, I have been imposing icosahedral symmetry and I was counting on doing symmetry expansion and focus classification (as used for SPA) for looking at the spike structure. I have expanded the symmetry using Relion but I am not able to do the particle subtraction part in order to define the smaller individual subvolume. Later, I would like to do 3D classification on subvolumes (spikes). I have tried Relion 3.1 and Relion 4.0 for the particle subtraction but it seems to not work on the 3D (input sub tomograms). I am wondering if someone had met that issue or has ideas for a pipeline that would allow me to get individual spikes from my virus reconstruction. I had to impose symmetry because of the low number of particles I can get (it is a rare biological process), so picking individual spikes instead of the whole virus is not an alternative that could work.
I have been using warp, IMOD, and Relion(3.1/4.0) for my virus reconstruction.
