I have a helical fibre complex that I'm fighting with a little bit with in RELION.
I have an accurate moderate resolution starting model made from the initial helical model process that visually makes sense and matches all the cross-over points and prior expectations. I know the unit repeat, the rise & the twist (accurate within a bit of an angstrom from 2D class averages and the layer lines of the FFTs), but my structure is popping out overly smooth and isnt quite right. There are 4 fibres twisiting around the long axis and I'm refining in C4, but I think i this is slightly wrong and the real symmetry is probably p1_4.
What is the convention for applying the pseudo symmetry in a reconstruction; I've been struggling to make sense of the documentation.
Any other tips /issues from users with more experience of helical refinements?
