I would argue that it is better to have no crystal in the loop as then there is always the possibility that the ones still in the drop that were not yet harvested might diffract!

I would argue that not getting diffraction would indicate that the sample under study is not a crystal ;-)
That way the two scenarios are the same - in neither case do you have a crystal in the loop
