My experiment has 2 task conditions and I would like to know whether the activity in an a priori ROI (amygdala) exhibits enhanced activation in condition A compared with condition B. If that is the case, I would further examine whether the functional connectivity from amygdala to other brain regions is also increased in condition A compared with condition B.
Before analyses, we defined an amygdala ROI from a relevant study (peak: x1/y1/z1; a sphere with r = 6). When we performed small-volume correction within this ROI, the result was significant, with a peak voxel at x2/y2/z2.
Then I want to extract the deconvolved time series of amygdala for PPI analysis. There are two options: (1) to extract time series from the a priori ROI, and (2) to extract time series from the suprathreshold peak x2/y2/z2, with a radius of, say, 3.
Anyone could suggest which option is better, or makes more sense?
