Hi Selina,
I think it should be the latter. You can find out in SPM.xX when you load SPM.mat into MATLAB workspace. SPM.xX.X is the design matrix and you can look into SPM.xX.name for reference to their name. If you use HRFs, then you may need to consider this when constructing your contrast. For their meaning, see this post:
