Dear colleagues,
We are studying data with two time points and two groups (n1=13 and n2=2). Following the VBM8 manual and Gläscher/Gitelman 2008 tutorial, our SPM8 flexible factorial design looks like the following:
- f1: subject, independence: yes, equal variance
- f2: group, independence: yes, unequal variance
- f3: time, independence: no, variance equal
- main effect for factor 1
- interaction for factors 2 and 3
With the following design matrix:
g1t1, g1t2, g2t1, g2t1, TIV, Site, Age, Subject1, Subject2 ... Subject 15

We were not sure how to define a proper contrast e.g. to obtain the group effects. Gläscher/Gitelman suggested to use the t-contrast:
0.5 0.5 -0.5 -0.5 0 0 0 1/13 1/13 1/13 1/13 1/13 1/13 1/13 1/13 1/13 1/13 1/13 1/13 1/13 -0.5 -0.5
However, SPM8 displays an 'invalid contrast' message, whatever we enter. Even a simple 1 1 -1 -1 is discarded.
Do we need to use an F contrast instead or do we have to modify the design? Any help is appreciated.
Best regards,
Martin
