A word of caution: not every macromolecular crystallography beamline will be able to determine the presence of Mg by XRF. Plus there may be Mg in the cryo-solution.
From a biochemical perspective: does the protein have Mg-binding sites? Recently a colleague of mine has proven that her protein has a binding site capable of binding both Mg and Mn by ITC and thermofluor melting assay and locate the binding site in a structure.
