I am acquiring longitudinal resting state fMRI in two groups undergoing two different treatments.
I have a really precise hypothesis, that is that connectivity between region X and Y will increase between time 0 and time 1 in subjects receiving treatment A but not treatment B.
To test this hypothesis I thought that the best way would be calculating a ROI to ROI connectivity. Meaning taking the time series from region X and Y from each subject in each time point, calculating the correlation between the two region and then test for an interaction between time and group.
My questions are:
1)is the preprocessing for doing ROI to ROI connectivity similar to the one you should use for seed-based connectivity (slice timing correction, BP filtering, realignment and movement correction, scrubbing and so on) ?
2)it is necessary to correct for WM and CSF time series when doing ROI to ROI connectivity ?
3)which toolbox I should use ? (I though about CONN).
4)have you any how to or tutorial and/or paper about ROI to ROI connectivity to suggest me ?
