Which duration you choose will depend on the interpretation you want
to give to the value.
The ordinary thing to do here would be to choose the duration of the
block (in seconds). Then marsbar will generate a predicted
time-course for a block of this length, and calculate the percentage
signal change from that predicted time-course. This gives you the
estimated signal change for one block.
