I am not aware of any such manual just for SPECT but if you need some help to get started, here is my 2 cents worth from very much a SPECT/PET SPM8 beginner:
1. From my understanding PET/SPECT/VBM is basically all a second level, group analysis.
2. If scans are in DICOM then convert to analyse/nifti using SPM “DICOM import” (or maybe MRIcron, which is very easy to use).
3. Use “Display” to set the origin at approximately the AC.
4. Use “Normalise (Est & Write)” to warp the scans to the SPM SPECT template (or your own template if you have one). Use the bounding box -90:91 -126:91 -72:109 so that the warped scans are the same size as the template.
5. Use “Smooth” to smooth the scans, I have been using the default 8x8x8 kernel.
6. Use “Basic models” to set up your model. I have been using a 2 sample t test to compare a group of patients to a control group, and a 1 sample t test to see the effect of age in each group (I think this is correct!?)
7. Use “Esimate” to run the model/calculate the stats.
8. Use “Results” to set up you contrasts and set your FWE/cluster thresholds.
9. Use “overlays” to visualise the blobs on a nice 3D surface render or the single_subj_T1 MRI.
I apologise if your question was more complicated than just how to get started.
The SPM8 manual is also a good start but I find it very fMRI centric.
If anybody knows of a good PET/SPECT SPM manual or similar reference please post the details.
