We do this a lot (if I've understood correctly):
Click on admin and you should see three columns: account, property and view (profile)
In the drop-down for the view you should see 'create new view'.
When you have created the basic view click on 'filters' to add a basic filter to just include the sub-directory.
We use this approach to ensure we can measure unique visitors to individual sections as well as the site overall (and have custom reports per section).
'Profiles' are now called 'views' as of very recently in case that's confusing things!
