Hi Di,
Although I am not the one who can explain something about microinjection,
(as I am not an expert), my grad students use 2-10 micrograms/milliliter
circular plasmid DNA for microinjection of the Ciona intestinalis embryo.
Reporter genes have been well expressed at these concentrations.
They sometimes prepare the plasmid solution using 1 micrograms/milliliter
of Fast Green.
