You may want to have a look at the Volumes Toolbox. The menu structure
is a bit confusing (for historical reasons), but you will find a helpful
utility under "Volume handling utilities"->"Single volumes"->"Extract
Data from ROI". Data source would be "Source images", and you can enter
one or more ROIs to extract from. Extracted data will be saved to MATLAB
workspace as a struct, containing raw voxel values, voxel/mm coordinates
sampled etc.
Also note that handling 4D data is in many cases slower than handling
3D, because it requires your OS to handle very large files even if you
only need to access a small portion of it.
