Sure; in SPM you could use ImCalc (one of the buttons in the top left
window), in the easiest case using "i1+i2". I wouldn't be surprised if
MRIcro could do this as well but am not sure; have you considered asking
that question on the MRIcro user support list

If you're talking PET, I am sure you are aware of the issues of changing
SNR with radioactive decay and weighting techniques for creating summed
images which would need to be considered depending on the application.
