Some more interesting films on cities are as follows:
The Man with the Movie Camera by Vertov (1925) - An elegy on Moscow life in the first few years after the October Revolution.
Berlin: The Symphony of a Great City by Ruttmann (1927)
Alphaville by Godard - A film noir based on a futuristic city
Wings of Desire by Wenders - A story of 'angels' strolling around Berlin. David Harvey wrote a chapter in his book 'The Condition of Postmodernity' about this film comparing/contrasting it with Blade Runner.
Chunking Express and Fallen Angels both by Wong Kar-wai. Stories in a nightmarish Hong Kong at night.
Amorres Perros by Inarritu - Parallel stories in Mexico City: the themes range from poverty and social inequality to terrorism.
At the Edge of the City by Giannaris (English Subtitles) - The film shows the life of young Greek-Russian refugees in Athens. Most of the film is shot at night.
Fate by Demirkubuz (English Subtitles) - The story is loosely based on Camus' "The Stranger" and shows the life of a middle-aged Turk in Istanbul who who has largely given up on the idea of free will and lives with no sense of direction, resigned to let life take its own course.

I am not sure if we could call it the first, but Fritz Lang’s 1927 classic Metropolis must be among the earliest urban-themed films. It’s an expressionist film dealing with class struggles in a futuristic urban setting. A silent film classic, it was restored and re-released in 1984 and 2001. A must for this list.
