I can't see any problem here. You can have entirely different
operating systems running in different virtual machines, e.g. Linux,
WinXP and Win95 if you really wanted it, so there should be no
difficulty in having WinXP with dll27 and WinXP with dll42. You could
do this kind of thing with dual booting as well, you'd just need to
partition the disk and have separate installations.
Regarding timing issues, have you looked at:

Obviously this won't answer any research questions about whether the
delays are significant, but it should help you work that out in your
particular case.
Regarding hardware, hopefully this link:

should point you to a FAQ answer within vmware, if it's broken just go
to vmware.com and search the "knowledge base" for hardware
compatibility. It sounds to me like most USB/serial/parallel connected
things should be accessible, but that unusual PCI cards probably won't
be, but I've not looked into this properly, and certainly haven't
tried for real.
