Dear Tibor:
I only work with 3-D images using SPM5. Here is one way to read 3-D
nifti images.
I use a function called "spm_vol.m"
A=spm_vol('name_of_file.img'); %Read an image file
3-D_Intensity=A.private.dat(:,:,:); %3-D data
