You have to reslice your images at least once between realignment and
statistics. If you normalise your images, then "normalisation -> write
normalised" will take care of the realignment information. Without
normalisation you need to do "realign -> coregister & reslice".
Deleting the .mat file will not only delete realignment information, but
also any information about slice orientation that was present in the DICOM
images (and which might be useful to coregister between different scans
with different slice positioning).
