As some those have replied have pointed out, you can't just reuse SPM.mat
if you're too far into processing.
For example, assuming you're using SPM2 (actually, which version are you
using?---that's important to answering the question), if all you've done
is press "data" and not "design" (I assume this is for fMRI not PET, and
for SPM2 not SPM99), then the matrix is completely resusable.
In principle, it should be pretty easy to extract the "design only" part
of SPM.mat. A small matlab script/function could be written to rip out
the stuff that's particular to one subject.
