Genetic analysis allowing for genotyping errors
Genotyping errors are known to reduce the power of genetic linkage and
association analyses. Unfortunately, detection of such errors is usually
difficult, and sometimes impossible (e.g. for case-control studies). Even when
errors are visible, removing the data from these families may bias the analysis.
Methods for performing analyses allowing for undetected genotyping errors are
presented for the following three areas:
1) Association analysis using unrelated cases and controls.
2) Family-based association analyses on cases and their parents.
3) Linkage analysis of affected sibling pairs.
The methods give tests with correct Type I error rate and approximately
unbiased estimates of risk parameters in the presence of genotyping errors.
However, the power of the tests still decreases as the amount of genotyping
error increases.
