I have tried a number of "liquidizers" in the past for homogenizing
shellfish. The thing to remember is the use of the homogenate. Many
methods of homogenization heat up the homogenate, and destroy its usefulness
for the analytical method. Here are a few suggestions:
1. There are manufactured mortar and pestle apparatus like those used in
biochemistry. One with a Teflon pestle works well for prepping hard
biological samples if you make a few modifications. Notch the pestle with a
file in a star pattern at the bottom. This will act like teeth and help to
grind the tough foot. Then use a drill and drill press to power the pestle,
and save some energy on your part. Another advantage of this method is that
you can place the test-tube-like mortar in an ice bath to keep the tissue
cool.
2. There are a number of commercial grinding apparatus on the market. These
devices work with two blades that are concentric to one another. The one
moving blade is called the rotor; the immobile one is called the stator. I
was able to grind snails and conchs using this method. Choose a brand that
has detachable cutters, and a choice of sizes for the cutters. Then you can
choose the most appropriate size for your sample volume. Buying more than
one blade can also speed the process if you are interested in avoiding
intersample contamination.
3. Sonicators work pretty well if you pre-cut (using a scalpel or scissors)
the sample into slightly smaller and more or less uniform chunks. These
sonicators also are available with different sized horns to accommodate your
sample size.
